SUMMARY:YCC Virtual April Seminar-How to Translate Your Academic Studies into an Industry Career
DESCRIPTION:Title:  How to Translate Your Academic Studies into an Industry Career \nSpeaker: Mufeda Zhort \nBio: Mufeda Zhort holds a bachelor’s in chemistry concentration in Pharmaceutical Science from Fairleigh Dickinson University\, NJ. She further pursued her master’s and Ph.D. in chemistry from Seton Hall University\, NJ\, where she specialized in the synthesis\, structure elucidation and alkane oxygenation reactivity of three bioinspired fluoroalkyl iron phthalocyanines. \nShe joined PDI in May 2020 where she worked in a cGMP lab as an analyst\, method validation and transfer\,  and as a data reviewer. She then joined Parexel (formally Execupharm) contracted at Merck in the Discovery Analytical R&D group collaborating with the high throughput experimentation and high throughput purification groups for method development of small and large molecules and was promoted to Principal Scientist 1 after her tenure. \nIn the beginning of this year\, Mufeda started her new role as a Senior Scientist at Merck in the Small Molecule Analytical Research and Development group at West Point\, PA. She is part of the GLP Analyical team developing innovative analytical methodologies for early stage development. \nJoin Zoom Meeting\nMeeting ID: 864 8599 8602\nOne tap mobile\n+16465588656\,\,86485998602# US (New York)\n+16469313860\,\,86485998602# US

SUMMARY:North Jersey NMR Topical Group
DESCRIPTION:Speaker: Boyd M. Goodson\, Associate Dean – Research\, College of Agricultural\, Life\, & Physical Sciences at Southern Illinois University \nTitle: Parahydrogen-Based Hyperpolarization Techniques for Spectroscopy and Imaging with New Contrast Agents and ‘Point-of-Care’ MRI Scanners \nAbstract: NMR and MRI enjoy wide applicability but suffer from poor detection sensitivity. For example\, MRI can provide exquisite anatomical images of soft tissues\, but the signal comes almost entirely from high-concentration 1H nuclei in the body’s water and fat molecules. It would be extremely informative to instead obtain MRI signals from various lower-concentration species. We are pursuing methods for generating high\, non-equilibrium nuclear spin polarization—hyperpolarization—that can transiently increase the sensitivity by orders of magnitude. One family of approaches uses parahydrogen to create hyperpolarization. These methods use catalysts to facilitate the hyperpolarization process–either by giving rise to hydrogenation of a substrate’s unsaturated chemical bonds (PHIP)\, or by having the catalyst reversibly bind the substrate and parahydrogen to allow hyperpolarization transfer via scalar couplings (i.e. SABRE). We are studying PHIP and SABRE with homogeneous and heterogeneous catalysts\, in aqueous environments\, and in variable magnetic fields to create hyperpolarized agents for NMR and MRI. We are interested in approaches that enable efficient separation of hyperpolarized molecules (both liquids and gases) from PHIP/SABRE catalysts\, and/or allow longer-lasting hyperpolarization—particularly in heteronuclei Finally\, we are working to integrate parahydrogen-based hyperpolarization with a new type of portable low-field (64 mT) “point-of-care” clinical MRI scanner. \n  \nThis will be a virtual meeting hosted via Microsoft Teams. \nJoin now

DESCRIPTION:Program:\n5:30 PM    Social and Registration\n6:15 PM    Complimentary Dinner\n6:55 PM    Welcome and Opening Remarks\n7:05 PM     Speaker 1\n8:00 PM     Speaker 2 \nFirst Speaker:  Xuanwen Shawn Li\, Principal Scientist at Merck & Co \nTitle: Identification and Control of High-Risk Host Cell Proteins for Biopharmaceutical Process Development with Advanced LC-MS-Based Proteomic Approaches \nAbstract: Host cell proteins (HCPs) are bioprocess impurities originating from the host cells used in biopharmaceutical manufacturing. Identifying and controlling high-risk HCPs is essential for maintaining product quality and shelf life. Notably\, HCPs with hydrolase activity can degrade formulation excipient polysorbates\, reducing drug stability. In this presentation\, we will discuss advanced LC-MS-based proteomic approaches\, including high-throughput proteomics and activity-based protein profiling (ABPP)\, to identify high-risk HCPs for biopharmaceutical process development and to enable product quality control. \n  \nSecond Speaker: Matt Willetts\,  US Proteomics Applications Manager at Bruker Scientific\, LLC \nTitle: New developments in Single Cell Proteomics and Immunopeptidomics on the timsTOF Ultra 2 \nAbstract: Recent advancements in the timsTOF Ultra 2 system have significantly impacted the fields of single-cell proteomics and immunopeptidomics. We will present new workflows for single cell analysis that enable more than 1000 single cells to be analyzed per day and new acquisition and data processing methods for immunopeptidomics. We will also present our new AIP technology which promises further improvements in peptide and protein IDs. \nSponsor:   \n \nPlease register.  It’s free\, including dinner\, but registration is required.

SUMMARY:North Jersey ACS NMR Topical Group
DESCRIPTION:Primary transfer step in the light-driven ion pump bacteriorhodopsin: An irreversible U-turn revealed by dynamic nuclear polarization-enhanced magic angle spinning NMR \nDr. QingZhe Ni\, Senior Scientist\, Merck \n  \nAbstract: Despite much attention\, the path of the highly consequential primary proton transfer in the light-driven ion pump bacteriorhodopsin (bR) remains mysterious. Here we use DNP-enhanced magic angle spinning (MAS) NMR to study critical elements of the active site just before the Schiff base (SB) deprotonates (in the L intermediate)\, immediately after the SB has deprotonated and Asp85 has become protonated (in the Mo intermediate)\, and just after the SB has reprotonated and Asp96 has deprotonated (in the N intermediate). An essential feature that made these experiments possible is the 75-fold signal enhancement through DNP. 15N(SB)–1H correlations reveal that the newly deprotonated SB is accepting a hydrogen bond from an alcohol and 13C–13C correlations show that Asp85 draws close to Thr89 before the primary proton transfer. Concurrently\, 15N–13C correlations between the SB and Asp85 show that helices C and G draw closer together just prior to the proton transfer and relax thereafter. Together\, these results indicate that Thr89 serves to relay the SB proton to Asp85 and that creating this pathway involves rapprochement between the C and G helices as well as chromophore torsion. \nJoin the MS TEAMS meeting
